Abstract Synopsis

  • 14-3-3 proteins interact with ligands that have phospho-serine motifs, but the details of how this binding and release occurs are not fully understood.
  • Two key acidic residues (D129 and E136) in the 14-3-3 protein's binding pocket influence its ability to form or break complexes with ligands like NPM1, affecting centrosome duplication.
  • Experimentally changing these residues to alanine revealed opposing effects on centrosome duplication, highlighting the important regulatory role of 14-3-3γ in this process and providing new insights into how these protein complexes form and dissociate.
