Abstract in brief

VEXAS syndrome is an autoinflammatory condition linked to somatic mutations in the UBA1 gene, presenting with diverse symptoms such as skin lesions, fever, weight loss, lung issues, ocular problems, relapsing chondritis, venous thrombosis, lymph node swelling, and joint pain.
In a study of 116 French patients, about half had hematological issues like myelodysplastic syndrome (MDS) and monoclonal gammopathy, with different UBA1 mutations associated with conditions and outcomes, showing a range of disease severity and prognosis.
The analysis identified three patient clusters with distinct clinical profiles and survival rates: mild to moderate disease, those with MDS and higher mortality, and others with systemic symptoms, with the UBA1 pMet41Leu mutation linked to a better outlook.
