Abstract Synopsis
- RNABloom2 is a new referencefree method for assembling longread transcriptome sequencing data, which can reconstruct transcripts without needing a reference genome, making it useful for studying species without high-quality genomes.
- The method performs comparably to existing referencebased tools in terms of assembly quality, while also using significantly less memory (270-806%) of the peak memory and less runtime (36-108%) than other referencefree methods, making it more efficient.
- RNABloom2 was successfully used to assemble the transcriptome of Picea sitchensis (Sitka spruce), demonstrating its potential for large-scale comparative transcriptomics in species lacking readily available genome references.]
