Abstract Synopsis

  • This research introduces a new method for preparing DNA libraries that enabled scientists to sequence a high-quality, 30x coverage genome of a Denisovan, an extinct relative of Neandertals, revealing very low genetic diversity in these ancient humans.
  • The high-quality genome data allows researchers to estimate that Denisovans had extremely low heterozygosity and to preliminarily date the specimen based on its genetic differences, helping to understand when they lived.
  • The study also provides detailed insights into how Denisovan and Neandertal DNA have mixed into modern humans and identifies key genetic changes that became common in Homo sapiens after diverging from Denisovans.]
